Shimano GRX 1x chainring size limitations.

Shimano only sell 40T and 42T, but can you put a 46T on?  Or will that cause issues with the default 11-42 cassette?

As long as you only have one chainring up front, it can be any size you want. You'll have to get a longer chain or do some quick link magic to make it work. Also, make sure a larger chainring will actually fit on your bike before you buy.

For a 1x setup, I don't see why not. The limiting factor would actually be your chainstay. Usually, if you got clearance for wider rear tire, the chainring clearance would have to give.
